Abstract

Ludwigia octovalvis termasuk salah satu jenis tumbuhan tropis yang berpotensi sebagai tumbuhan obat. Bioaktivitasnya sebagai antidiabetes dalam menghambat enzim α-glukosidase dari bagian buah tumbuhan ini belum pernah dilaporkan begitu juga dengan metabolit sekundernya. Penelitian ini bertujuan untuk mengisolasi senyawa metabolit sekunder dan penentuan aktivitas antidiabetes dalam menghambat enzim α-glukosidase dari ekstrak metanol, fraksi n-heksana, fraksi etil asetat dan senyawa hasil isolasi. Proses ekstraksi dilakukan dengan cara maserasi dengan menggunakan pelarut metanol, kemudian dilanjutkan dengan fraksinasi cair-cair menggunakan pelarut n-heksana lalu dilanjutkan dengan pelarut etil asetat. Proses pemisahan dan pemurnian senyawa dilakukan dengan menggunakan kromatografi cair vakum (KCV), kromatografi radial dan kromatografi kolom flash. Senyawa hasil isolasi yang diperoleh berupa padatan putih (86,2 mg) dengan titik leleh 193,5оC-194,8оC. Analisis struktur senyawa hasil isolasi menggunakan spektroskopi UV dan NMR (1H-NMR dan 13C-NMR) menunjukkan bahwa senyawa hasil isolasi adalah senyawa metil galat dan baru pertama kali dilaporkan dari tumbuhan L. octovalvis. Aktivitas penghambatan enzim α-glukosidase menghasilkan nilai IC50 dari ekstrak metanol, fraksi n-heksana, fraksi etil asetat, dan senyawa hasil isolasi secara berturut-turut adalah sebesar 8,6; 167,8; 3,0 dan 139,7 ppm dibandingkan dengan kontrol positif akarbosa dengan nilai (IC50 129,8 ppm). Berdasarkan hal ini buah L. octovalvis berpotensi sebagai antidiabetes. Bioactivity as antiidiabetic in the α-glukosidase inhibiton assay as well as the secondary metabolite from the fruit L. octovalvis have never been reported. The research was conducted to isolate secondary metabolites and determine antidiabetic activity in inhibiting the α-glucosidase enzyme from methanol extract, n-hexane fraction, ethyl acetate fraction and the isolated compounds. The extraction process was done by maceration using methanol solvent, then continued with liquid fractionation sequentially using n-hexane and ethyl acetate solvent. The process of separation and purification of compounds were carried out using vacuum liquid chromatography (VLC), radial chromatography and flash column chromatography. The isolate compound was a white solid (86.2 mg) with a melting point 193.5оC-194.8оC. The structure analysis of the isolated compound using UV and NMR (1H-NMR and 13C-NMR) spectroscopy showed that the isolated compound was a phenolic group compound derived from gallic acid namely methyl gallate with an other name methyl-(3,4,5-trihydroxybenzoate). Based on literature studied, the compound was reported for the first time from this species. The IC50 of inhibiting α-glucosidase enzyme activities from methanol extract, n-hexane fraction, ethyl acetate fraction, and the isolated compound were 8.6, 167.8, 3.0 and 139.7 ppm which were compared with the positive control of acarbose (IC50 129.8 ppm).
