Abstract
Ajwa dates contain flavonoid compounds that have antioxidant activity that can inhibit and bind reactions from free radicals. The purpose of this study was to determine optimization of formula granule of ethanol extract ajwa dates that are practical, consumable and have antioxidant activity. The granule dosage form was chosen because this dosage form is more stable if stored for a long time, has the right dosage and is easy to prepare when it will be consumed. The ethanol extract granules of ajwa date palm flesh are made in nine formulas with PVP variations as a binder and explotab as a disintegrant to determine the optimum formula characteristics of the granules to be optimized by factorial design methods. Ethanol extract of ajwa dates was obtained by maceration method and characterization and phytochemical screening were carried out. The test response in determining the optimum formula of the granule includes moisture content, granule flow speed, rest angle, compressibility, and soluble time so that the optimum formula of the granule is obtained. The optimum formula was then tested for antioxidant activity with DPPH and stability test for 28 days in a 25°C oven. The total flavonoid content in the ethanol extract of ajwa date palm flesh was 112,25 ± 0,03 mgQE/g of extract. The optimum formula of the granule was obtained at a PVP concentration of 3,5% and an explotab of 8%. The results of the antioxidant activity test with DPPH showed that the optimum formula of the granule has an IC50 of 60,034 µg/ml which has a strong category of antioxidant activity. The results of the granule optimum formula stability test showed no significant changes in the test parameters of granule flow speed, angle of rest, compressibility, and soluble time on day 1 to day 28 as well as a significant change in moisture content on day 21 to day 28. Based on the response analysis using the factorial design carried out, it can be concluded that pvp variations have a significant effect on determining the optimum formula produced.
