Abstract

MIFTAHUL JANNA. In-vitro Analysis of Anti-hypercholesterolemia Activity of Polyphenol and Flavonoid Compounds Water Lettuce (Pistia stratiotes) Leaf (supervised by Sabri Sudirman). The enzyme 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase is an enzyme that plays a role in the synthesis of cholesterol. Synthetic anti-cholesterol drugs such as statins have been used as HMG-CoA reductase inhibitors. However, these drugs have side effects, so natural HMG-CoA reductase inhibitors such as from plants origin are needed. This study aimed to determine the in vitro potential inhibitory HMG-CoA reductase activity as an anti-hypercholesterolemia property of the water lettuce (Pistia stratiotes) leaf extracts. The research was conducted experimentally in a laboratory with a treatment consisting of 2 levels, namely crude extract and purified extract and repeated 3 times. Total polyphenols, flavonoids, and inhibitory activity of the HMG-CoA reductase assay were carried out by in vitro analysis. The results obtained were analyzed quantitatively, followed by using different parametric statistics (Independent sample t-test), and presented in graphical form. The results of this study showed that the yield of the extract was 4.13% (crude extract) and 3.34% (purified extract). The total polyphenols in each extract were 29.03 mg GAE/g dry sample (crude extract) and 65.63 mg GAE/g dry sample (purified extract). The total flavonoids were 27.58 mg QE/g dry sample (crude extract) and 88.02 mg QE/g dry sample (purified extract). The inhibitory activity of the HMG-CoA reductase enzyme showed that the purified extract could inhibit (34.74%) higher than the crude extract (2.61%). The purified extract of water lettuce has higher levels of polyphenols and flavonoids and can inhibits the HMG-CoA reductase enzyme more effectively than crude extract.
