Abstract

Durian (Durio zibethinus Murr.) is a tropical fruit plant native to mainland Southeast Asia. Plant tissue culture is a plant propagation technology from cells, tissues and plant organs in solid or liquid media under aseptic conditions. This study was aimed to evaluate the effectiveness of several sterilization methods, as well as to obtain the best sterilization procedure for durian leaf midrib explants in tissue culture activities with the addition of sodium hypochlorite compounds. This research was carried out at the Tissue Culture Laboratory, Faculty of Agriculture, Sriwijaya University, Bukit Lama, Ilir Barat I District, Palembang City, South Sumatra with location coordinates 2o59'23.4''S 104o43'53.4''E. The research period started from June to September 2024. This sterilization treatment used a combination of sterilant ingredients, namely 70% alcohol, bactericide, fungicide, liquid detergent, and NaOCI. Data presentation used parametric descriptive analysis by calculating the percentage of live explants, contaminated explants and contaminated media. The results of this research were the sterilant combination treatment 5(P5), namely liquid detergent for 3 minutes, 0.2% bactericide for 10 minutes, 0.2% fungicide for 10 minutes, 1.5% NaOCI for 5 minutes and 70% alcohol for 5 minutes (54%) showed the best result and corsidered more effective (54%) with the highest percentage of live explants and a fairly low percentage of explant contamination compared to other treatments.
