Abstract

Pancreatic lipase is an enzyme that plays a role in the absorption of triglycerides in the human body. Performance that continues to increase triggers the activity of fat accumulation in adipose tissue so that there is a risk of being overweight or obesity. The drug orlistat is often used to inhibit pancreatic lipase activity, but this drug can have adverse reactions to health. Based on this, natural pancreatic lipase enzyme inhibitors from plant bioactive compounds are needed. This study aims to measure and examine the potential of pancreatic lipase enzyme inhibitory activity as an anti-hyperlipidemia by crude extract and extract purification of polyphenolic compounds from the leaves of apu-apu (Pistia stratiotes) in vitro. The research was conducted experimentally in a laboratory consisting of 2 levels (crude extract and purified extract) carried out 2 (times) replicates, using a qualitative test of polyphenol compounds and using an in vitro lipase enzyme inhibitory activity test. The data obtained were analyzed descriptively in the form of tables and graphs, followed by parametric statistical analysis of the different t-test (Independent Sample t-Test). The results showed that the crude extract yield was 1.57 g/100 g sample, purified extract was 0.81% g/100 g sample. Phytochemical qualitative test showed that both extracts contained flavonoid compounds, tannins, and terpenoids. and The inhibitory activity of pancreatic lipase enzyme by crude extract was 37.50%, purified extract was 72.50%, and pravastatin as positive control was 95.66%. Based on the data, it was found that the best potency as an inhibitor of pancreatic lipase enzyme was possessed by purified extracts.
